DNA transfection:
A wide variety of mammalian cells can be transfected with SAINT-MIX.
SAINT-MIX has been used to transfect several cell-types/lines, some with efficiencies up to 100%.

Transfection in large cultures vessels and roller bottles is expensive in labor and media
needed for the culturing of the cells. Finally the general outcome of the transfection
experiments shows only 10-15% of the input cells are transfected and massive cell-death is
induced due to the transfection protocol.

SAINT-MIX is an efficient transfection device which transfect all kind of cell-lines.
The cell doubling time of cells and the incubation time of the SAINT-MIX/ DNA complexes on the cells are factors which can influence the transfection efficiency. Previous studies of time courses showed that incubation of the cationic lipid/ DNA complexes is an important factor which affects the transfection efficiency.

SAINT -MIX proved to be a very efficient transfection system for the transfection of several eukaryotic cell lines (see references) we undertook to investigate whether SAINT_MIX could be successfully used to transfect ES cells.

Oligonucleotides:
SAINT-MIX has been demonstrated to act as an efficient translocator
of oligo-nucleotides into eukaryotic cells. The compatibility with serum is a major advantage of this delivery system. Oligonucleotide induced cell-damage will be minimal.

The SAINT-MIX delivery system is based on a cationic pyridinium head group, showing excellent bio-compatibility.
Molecular structure of SAINT-18 (1-methyl-4-(cis-9-dioleyl)
methyl-pyridinium-chlorid):

SAINT-MIX consists of SAINT and DOPE (Dioleoyl-phosphatidylethanolamine) dissolved in H2O. The SAINT-18: DOPE ratio is 1:1.
Stability experiments showed that SAINT-MIX, stored at 4°-7°C, is stable for at least one year after opening. No loss of function appeared after repeatedly opening of the vials as long as the vials are recapped tightly after use. The antibiotic properties of SAINT-MIX ensure sterility for prolonged periods.
