1ml SAINT-RED is sufficient to perfrom 600 tfx on a 24 well format with 4 pmol siRNA.
- Inhibition of EpCAM expression on SKOV-3 tumour cells.
We compared several siRNA transfection reagents by evaluating the expression inhibition of the epithelial-glycoprotein EpCAM on SKOV-3 tumor cells. The results clearly show that SAINT-RED mediated delivery of siRNA generates the highest inhibition by far compared to the competitors using only 3 pmol of siRNA.

Synvolux Therapeutics siRNA transfection reagent SAINT-RED permits the rapid and efficient delivery of siRNA into a variety of mammalian cell lines. Here, the application of SAINT-RED in siRNA high-throughput screening is demonstrated. It also has been shown that both the forward and reverse transfection protocol yield similar results.
- Inhibition of enzyme activity with different amounts of siRNA.
150,000 3T3 cells were seeded in a 6-well plate the day before transfection.
Enzyme activity of a housekeeping gen was determined 48 hr after a single transfection of different amounts of siRNA. When 1 ug of siRNA (70 pmol) was complexed with 20 ul SAINT-RED an inhibition of 75% was found. Final concentration of siRNA is 25nM.
- Long term preservation and high transfection efficacy of siRNA.
Long term preservation and high transfection efficacy of siRNA is achieved by use of SAINT-RED. Reproducibility and the introduction of an (internal) standard is difficult when using siRNA to knock-down gene-expression. If the experiment does have a time-spanning period (several transfections in days/months), and contains different subsets of data to be generated, it is even more complex. The siRNA could become inactive even if you aliquot the solution and store at -20°C.
